@article{ธนานันต์_หงษ์ทองดี_ธนานันต์_2016, place={Pathumthani, Thailand}, title={การจำแนกชนิดและการประเมินความสัมพันธ์ทางพันธุกรรมของกล้วยไม้สกุลเอื้องเทียนด้วยเครื่องหมายแฮตอาร์เอพีดี}, volume={5}, url={https://li01.tci-thaijo.org/index.php/tjst/article/view/56896}, DOI={10.14456/tjst.2016.2}, abstractNote={<p><strong>บทคัดย่อ</strong><strong></strong></p><p>กล้วยไม้สกุลเอื้องเทียนพบได้ตามป่าลุ่มต่ำเขตร้อนและป่าดงดิบ เป็นกล้วยไม้ที่มีดอกสีสันสวยงาม มีกลิ่นหอมอ่อน ๆ ดูแลเพาะเลี้ยงง่าย และทนต่อความแห้งแล้ง จึงได้รับความนิยมในการปลูกเป็นไม้ดอกไม้ประดับ ปัจจุบันการจำแนกพันธุ์โดยดูเพียงลักษณะสัณฐานทำได้ยากและมีความผิดพลาดสูง ดังนั้นผู้วิจัยจึงศึกษาการจำแนกชนิดและประเมินความสัมพันธ์ทางพันธุกรรมของกล้วยไม้สกุลเอื้องเทียนด้วยเทคนิคแฮตอาร์เอพีดี โดยใช้ไพรเมอร์แบบสุ่ม 72 ไพรเมอร์ ผลการวิจัยพบว่า 37 ไพรเมอร์ สามารถเพิ่มปริมาณดีเอ็นเอของเอื้องเทียนทั้ง 14 ชนิด ได้ หลังจากนั้นได้คัดเลือกมา 31 ไพรเมอร์ เพื่อใช้ในการสร้างลายพิมพ์ดีเอ็นเอ ซึ่งปรากฏว่าสามารถแยกความแตกต่างระหว่างชนิดได้ด้วยแถบดีเอ็นเอที่จำเพาะ โดยพบแถบดีเอ็นเอที่ให้ความแตกต่างทั้งหมด 257 แถบ นอกจากนี้ยังพบไพรเมอร์ที่สามารถจำแนกเอื้องเทียนแต่ละชนิดออกจากกันได้ด้วยการใช้ไพรเมอร์เพียงชนิดเดียว จำนวน 10 ไพรเมอร์ เมื่อวิเคราะห์ด้วยโปรแกรม NTSYS และสร้างแผนภูมิความสัมพันธ์ด้วยวิธีการจัดกลุ่มแบบ UPGMA พบว่ามีค่าสัมประสิทธิ์ความเหมือนระหว่าง 0.31 ถึง 0.64 และสามารถจำแนกกล้วยไม้สกุลเอื้องเทียนออกเป็น 5 กลุ่ม ซึ่งผลการวิจัยนี้สามารถนำไปใช้ในการวางแผนเพื่อการอนุรักษ์และการปรับปรุงพันธุ์ต่อไป</p><p><strong>คำสำคัญ :</strong> กล้วยไม้; สกุลเอื้องเทียน; ความสัมพันธ์ทางพันธุกรรม; การจำแนก; แฮตอาร์เอพีดี</p><p> </p><p><strong>Abstract</strong></p><p>The <em>Coelogyne</em> can be found from tropical lowland forests to montage rainforests. The plants have beautiful, fragrant flowers, can tolerate drought and neglect. So, <em>Coelogyne</em> are popular orchids of growers by dwellings. Nowadays identification based on morphology is difficult and high error. Thus, High annealing temperature-random amplified polymorphic DNA (HAT-RAPD) technique was used to identify and assessment 14 species of <em>Coelogyne</em>. The total 72 random primers were screened and 37 primers could be used for DNA amplification. And then, selected thirty-one primers that can gave clear amplified products for used to construct DNA fingerprints. The result showed differences among 14 samples with specific DNA bands and were 257 polymorphic bands. In addition, 10 of 31 random primers were found to identify each sample using only one primer. A dendrogram, which constructed base on polymorphic bands using the NTSYS program, showed genetic similarities among the 14 species of <em>Coelogyne</em> with similarity coefficients ranging from 0.31 to 0.64 and separated to 5 clusters.
