VL - 8 IS - 3 SE - วิทยาศาสตร์ชีวภาพ DO - 10.14456/tjst.2019.35 UR - https://li01.tci-thaijo.org/index.php/tjst/article/view/186047 SP - 258-270 AB - <p><strong>Abstract</strong></p><p>Nucleotide sequences of nrITS were analyzed in order to evaluate genetic variation in 14 Thai waterlilies, <em>Nymphaea</em> spp. using BioEdit program. The ITS (internal transcribed spacer) region of approximately 900 bp contains sequences of partial <em>18S</em> gene, ITS1, <em>5.8S </em>gene, ITS2 and partial <em>26S</em> gene. Nucleotide sequence variations at ITS1 and ITS2 were observed more than those at <em>18S, 5.8S</em> and <em>26S </em>genes. Comparison to that of ITS2, nucleotide length of ITS1 was longer, but GC contents were less. The <em>5.8S</em> gene sequence was 156 bp in all 14 waterlily samples. Maximum likelihood (ML) phylogenetic tree of 14 waterlilies was constructed based on nrITS sequences using MEGA6 program with 1,000 replications (bootstrap). The tree could classify 14 waterlilies into 5 groups of different subgenus, i.e. subgenus <em>Anecphya</em>, subgenus<em> Brachyceras</em>, Intersubgeneric hybrids (ISG) of subgenus<em> Nymphaea and</em> subgenus<em> Brachyceras</em>, subgenus<em> Lotos</em> and subgenus<em> Nymphaea</em>.
