Abstract

Gold labeling lgG flow (GLIFT) was developed to be the tool for detecting potato virus Y (PVY) in potato. The excellent property of PVY-GLIFT kit are accuracy, easy, convenient and rapid detecting for diagnostic and screening of potato virus free seed production in 5 minutes which is faster than ELISA test that has taken 4-5 hours. It was developed by the combination of the two technologies namely serology and lateral flow technique. The concentration of 1 mg per ml of protein in lgG from PVY which was immunized in rabbit was conjugated with colloidal gold. The gold particles are stable, easy to conjugate with lgG and very clear in reaction of red colour at test line and control line. The optimum amount of gold labeling lgG of PVY in the reaction is about 2 µl/cm which shows the better reaction than using 1.0 and 1.5 µl/cm. The appropriate amount of lgG of PVY for preparing test line is 1.5 µl/cm. Goat anti-rabbit gave the strong red colour on control line the same as goat ant mouse. The GLIFT kit is a commercial cheap tool which is convenient to carry out for detecting PVY in the potato field more than NCM-ELISA technique.
