Detection of Chrysanthemum stunt viroid (CSVd) from naturally infected Chrysanthemum in Wang Nam Khiao District

Abstract

Chrysanthemum samples showing stunting, chlorotic leaves, small and abnormal flowers collected by sampling in Wang Nam Khiao district in Nakhon Ratchasima province, all 23 collected chrysanthemum samples were determined for Chrysanthemum stunt viroid (CSVd) by reverse transcription – polymerase chain reaction (RT-PCR) technique. The 8 of 23 RT-PCR products were approximately 360 nucleotides in size. After cloning and sequencing, all the 8 RT-PCR products were 354 nucleotides in size. To compare with CSVd in GenBank database and analyze by Phylogenetic Tree, all of them were Chrysanthemum stunt viroid and 98-100 % identity with CSVd Accession Nos. AF394452, JQ685739, HQ891018, JX909290, JF414238, JF398538, Z68210, DQ094298, FN646407 and U82445.   By bioassay, tomato cv.Rutgers and chrysanthemum plants were inoculated with CSVd. Rutgers tomato displayed symptomless while chrysanthemum plants showed chlorosis and small leaves. All these bioassay plants were determined by RT-PCR for CSVd infection. This is the first report of CSVd natural infection of chrysanthemum in Thailand.
