Efficiency for Shoot Multiplication of ‘Srivijaya’ New Hybrid Pineapple Variety Using Three Tissue Culture Systems

Abstract

Pineapple is an economic crop in the world containing high nutrition. There are many pineapple varieties grown in the world but most varieties still be clones in Cayenne or Queen groups. New pineapple hybrid variety, ‘Srivijaya’, was bred. This variety showed superiority for fruit quality and resistant to disease. The planting materials of the ‘Srivijaya’ variety are still limited for farmers. Tissue culture technique provides for mass shoot production in short time. This research, ‘Srivijaya’ pineapple tissue culture was invested. Suckers were sterilized using alcohol 70 % for 5 minutes and NaOCl 6 % w/w with 3 drops of tween for 35 minutes. The sucker was cut into 4 pieces and culture to induce shoots. Shoot multiplication was studied on three different in vitro culture systems; solid, liquid and bioreactor. Completely randomized design was used with 4 treatments. They were 1/2 MS+BA 0,5 mg/L, 1/2 MS+BA 1 mg/L, 1/2 MS+BA 2 mg/L, and MS+BA 2 mg/L. with 4 replications. Each experimental unit contained 5 explants in solid and liquid culture and 10 explants for bioreactor. Total number of shoots, height and, classified number of shoots based on height (<1 cm, 1-1.5 cm, >1.5 cm) were measured after cultured 30 days. The results were analyzed by Analysis of Variance and DMRT was used to mean different analysis. The results showed that bioreactor was the most efficient in shoot multiplication of ‘Srivijaya’ hybrid pineapple giving 124.5 shoots compare to 30.25 and 34.75 shoots in solid and liquid cultures respectively.
