Differences of Media Concentration for In Vitro Chrysanthemum Conservation

Abstract

The in vitro conservation of chrysanthemum lines for breeding program and commercial micro-propagation purpose spend lot of budget, time and labor. The price of some chemicals component of media is quite high.  Therefore, this study was aimed to study the growth of chrysanthemum on MS medium at different concentrations. This experiment was arranged with factorial design in CRD including 2 main factors; 10 chrysanthemum lines (M14–1, M10/1, M14–7, MK38, M11–3, M10, M7–7, M14–9, M7–10 and M9–5) and 4 different concentration level of MS media, including with (MS (normal condition),  1/2 MS, 1/3  MS and  1/4  MS).   The results showed that plant height, leafs number, leafs length and leafs width of chrysanthemum at 4 weeks cultured in 1 /2 MS, 1/3 MS and 1/4 MS were not significantly different from the chrysanthemum cultured in normal MS. It indicated that the conservation method for chrysanthemum lines might be used the low media concentrations. However, the response of each lines was highly significantly different for plant height leafs number, leafs length and leafs width (P < 0.05).
