Rice Improvement by Anther Culture of F1 Hybrid between Lemont and Khao Dawk Mali 105

The panicles of the F1 hybrid rice (Lemont/Khao Dawk mali 105) of which the distance between the auricles of the flag leaf and the subtending leaf was 6-12 cm were selected for anther culture. These spikelets had anthers bearing the mid-uninucleate microspores. High percentage of callus formation was obtained when anthers were cultured on the N6 medium supplemented with 500 mg/l casein hydrolysate, 2 mg/l NAA, 1 mg/1 kinetin and 50 g/l maltose. The calli could regenerate into plantlets at high frequency when cultured on the MS medium containing 15% coconut water, 300 mg/l casein hydrolysate, 20 g/l sucrose, 2 mg/lBAP and 0.2 mg/l NAA. About forty percent and forty-two percent of double haploid plants were obtained when the regenerants were transferred on the MS medium containing 1 g/l yeast extract, 15% coconut water, 3 mg/ 1 BAP and 0.01% colchicines for 3 and 5 days, respectively.
