Abstract

Tissue culture techniques for propagation and conservating of Wai Takra Thong ( Calamus caesius Blume.) has been carried out at the Royal Chitralada Projects and Department of Horticulture, Kasetsart University. Attempts have been made to induce callus and plant regeneration from immature embryo of Wai Takra Thong. The immature embryos were cultured on the MS (Marashige and Skoog, 1962) medium to obtain clean culture of explants. Cut the explants into thin pieces and culture on MS, Y3 (Eeuwens, 1976) and WPM (Lloyd McCown, 1981) media additional with various concentrations of 2,4-D and BAP in plastic well plate. The culture on both MS media with 0.5 mg/l 2,4-D + 2 mg/l BAP and 0.1 mg/l 2,4-D + 4 mg/l BAP produced callus and multiplication. Shoot and root formation appeared on the same media.
