Species Identification of Marine Crabs in Ao Yang, Chanthaburi Province Using DNA Barcoding

Abstract
This study aimed to study the species identification of marine crabs in Ao Yang, Chanthaburi Province, using DNA barcoding. Based on nucleotide sequence analysis of the cytochrome c oxidase I (COI) gene in mitochondria of 57 marine crab samples, the COI gene region was successfully amplified by PCR, and nucleotide sequence data were obtained from all samples with the lengths of approximately 700 base pairs. When comparing the genetic data of the crab samples with genetic code information in the GenBank database, ten species of marine crabs from five families were identified, with genetic similarity values ranging from 98.05% to 100%. The species identification derived from DNA barcoding were consistent with morphological identification. The results of Neighbor-joining phylogenetic tree demonstrated that all crab samples were clustered into a monophyletic clade, with clear separation of each crab family (Portunidae, Menippidae, Calappidea, Eriphiidae, and Xanthidae) and each crab species along the phylogenetic lineage. This study suggests that the DNA barcoding is suitable and effective for verifying the species of marine crabs in Ao Yang, Chanthaburi Province, which is a representative of the eastern coast of Thailand, especially for some crab species with high morphological similarity or intraspecific morphological variation. The obtained data can be used as a future genetic reference database for marine crabs, supporting molecular taxonomic studies, identifying crab species for marine and coastal resource conservation, and verifying species accuracy in seafood products.
