Development and Evaluation of Antibacterial Activity against Streptococcus mutans of Mouthwash with Jatu-Phala-Tiga Herbal Formulation Extract

Abstract
The Jatu-Phala-Tiga herbal formulation has been traditionally used to relieve fever, relieve flatulence, treat eye diseases, restore the body’s constitution, restore digestive balance, inhibit microorganisms and suppress inflammation. However, its inhibitory effect against the growth of Streptococcus mutans, the primary causative agent of dental caries, has not yet been investigated. This study aimed to determine the total phenolic and flavonoid contents of the ethanolic extract of Jatu-Phala-Tiga, to evaluate its antibacterial activity against S. mutans, and to further develop the extract into Jatu-Phala-Tiga herbal mouthwash formulas. The results revealed that the Jatu-Phala-Tiga extract had total phenolic and flavonoid contents of 368.62 ± 5.15 mg gallic acid equivalent/g dry extract and 652.33 ± 6.8 mg quercetin equivalent/g dry extract, respectively. It also exhibited antibacterial activity against S. mutans with the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of 12.5 ± 0.00 and 200 ± 0.00 mg/mL, respectively and inhibited its biofilm formation. In addition, the Jatu-Phala-Tiga herbal mouthwash formula 3 (containing the extract with a concentration of 2MIC as an ingredient) displayed the highest antibacterial activity across all tested time intervals.
