การวิเคราะห์ความสัมพันธ์ต่อลักษณะต้านทานโรคเหี่ยวเขียวในมะเขือเทศพันธุ์ปลูกด้วยวิธี Association Mapping

บทคัดย่อ

Association mapping ที่อาศัยความสัมพันธ์ทาง linkage disequilibrium ในประชากรที่มีความหลากหลายสามารถให้ความละเอียดและความแม่นยำที่ดีขึ้นในการวิเคราะห์ความสัมพันธ์ระหว่างยีนหรือเครื่องหมายดีเอ็นเอต่อลักษณะทางฟีโนไทป์ที่ต้องการศึกษา เมื่อเปรียบเทียบกับการวิเคราะห์ความสัมพันธ์แบบลิงค์เกจ ดังนั้น การศึกษานี้จึงมีวัตถุประสงค์เพื่อระบุตำแหน่ง (Quantitative trait loci; QTL) ต้านทานโรคเหี่ยวเขียวในมะเขือเทศด้วยวิธี association mapping โดยใช้ positional candidate loci และ functional candidate gene มะเขือเทศทั้งหมด 92 สายพันธุ์ ประกอบด้วย พันธุ์ต้านทานจากนานาชาติ 51 สายพันธุ์ พันธุ์การค้าของไทย 2 สายพันธุ์ และมะเขือเทศอ่อนแอ 39 สายพันธุ์ ถูกใช้สำหรับการวิเคราะห์ความสัมพันธ์ต่อลักษณะที่ศึกษาด้วยวิธี general linear mode โดยโปรแกรม TASSEL ความสัมพันธ์ระหว่างเครื่องหมายดีเอ็นเอต่อลักษณะที่ศึกษาถูกพิจารณาจากค่า probability (P) ที่ P < 0.05 ผลการวิเคราะห์ พบว่า เครื่องหมายดีเอ็นเอที่แสดงความสัมพันธ์กับความต้านทานโรคเหี่ยวเขียวส่วนใหญ่เป็นตำแหน่ง (QTL) ที่เคยมีรายงานไว้แล้ว ได้แก่ TG48 (โครโมโซม 2), TG118 และ CP18 (โครโมโซม 6) และ T0989 (โครโมโซม 12) นอกจากนี้ ตำแหน่งใหม่บนโครโมโซม 1 ที่มีความสัมพันธ์ระดับปานกลางกับความต้านทานโรคเหี่ยวเขียวถูกตรวจพบสำหรับเครื่องหมายพันธุกรรม SSR134, SSR572, SSR117 และ SSR65 สำหรับการวิเคราะห์ความสัมพันธ์ด้วย functional candidate gene พบว่า ยีนต้านทานโรค 4 ยีน คือ LRR, ascorbate peroxidase, jasmonate และ ethylene มีความสัมพันธ์ต่อลักษณะต้านทานโรคเหี่ยวเขียว ดังนั้น การศึกษานี้ยืนยันได้ว่าการวิเคราะห์ความสัมพันธ์ต่อลักษณะโดยวิธี association mapping เป็นอีกวิธีการหนึ่งที่สามารถใช้ตรวจหาตำแหน่งที่สัมพันธ์ต่อลักษณะที่ต้องการศึกษาได้เช่นเดียวกับการวิเคราะห์แบบลิงค์เกจใน ประชากรที่เป็น bi–parental
