การขยายพันธุ์ฟิโลเดนดรอนเบอร์กิ้น: อิทธิพลของสารควบคุมการเจริญเติบโตของพืชไบโอรีแอคเตอร์จมชั่วคราวและวัสดุปลูก

บทคัดย่อ

การศึกษานี้มีวัตถุประสงค์เพื่อพัฒนาวิธีการเพาะเลี้ยงเนื้อเยื่อเพื่อขยายพันธุ์ฟิโลเดนดรอนเบอร์กิ้น (Philodendron 'Birkin') โดยศึกษาผลของสูตรอาหาร สารควบคุมการเจริญเติบโตของพืช ระบบไบโอรีแอคเตอร์จมชั่วคราว (TIB) และการย้ายปลูก ยอดอ่อนที่เพาะเลี้ยงบนอาหารสูตร MS, WPM และ DKW ที่เติม Kinetin 0.5 มิลลิกรัมต่อลิตร เป็นเวลา 8 สัปดาห์ พบว่าไม่มีความแตกต่างอย่างมีนัยสำคัญระหว่างสูตรอาหาร โดยอาหารสูตร MS ให้จำนวนยอดเฉลี่ย 1.50 ยอด การศึกษาผลของไซโทไคนิน พบว่า BAP มีประสิทธิภาพในการเพิ่มจำนวนยอดสูงกว่า Kinetin อาหาร MS ที่เติม BAP 2.0 มิลลิกรัมต่อลิตร ให้จำนวนยอดเฉลี่ยสูงสุด (36.40 ยอด) ซึ่งสูงกว่าชุดควบคุมที่ไม่เติมสารควบคุมการเจริญเติบโต 22.8 เท่า อย่างไรก็ตาม BAP ความเข้มข้นสูงมีผลยับยั้งการเกิดราก การเติม IBA 1.0 มิลลิกรัมต่อลิตร ร่วมกับ BAP ยับยั้งการเพิ่มจำนวนยอดและชักนำให้เกิดแคลลัส ยอดที่ไม่มีรากสามารถพัฒนารากได้บนอาหารสูตร MS ที่ไม่เติมสารควบคุมการเจริญเติบโต ผลการศึกษาเบื้องต้นพบว่ายอดที่เพาะเลี้ยงในระบบ TIB เป็นเวลา 4 สัปดาห์ ให้จำนวนยอดเฉลี่ย 2.33 ยอด และความยาวยอด 3.37 เซนติเมตร สูงกว่าการเพาะเลี้ยงบนอาหารกึ่งแข็งเป็นเวลา 8 สัปดาห์ (1.50 ยอด และ 2.20 เซนติเมตร) แสดงให้เห็นว่าระบบ TIB ส่งเสริมการเจริญเติบโตได้ดีในระยะเวลาที่สั้นกว่า ในการย้ายปลูกต้นกล้ามีอัตราการรอดชีวิต 100 เปอร์เซ็นต์ โดยวัสดุปลูกผสมพีทมอสและขุยมะพร้าว (1:1) ให้ผลดีที่สุดในด้านจำนวนใบ ความสูงต้น และการพัฒนาราก ผลการศึกษานี้แสดงถึงศักยภาพของเทคนิคการเพาะเลี้ยงเนื้อเยื่อพืชสำหรับการขยายพันธุ์ฟิโลเดนดรอนเบอร์กิ้นในเชิงพาณิชย์
