In Vitro Preservation of Vanda coerulea Giff. Protocorm-Like Bodies

Protocorm-like bodies (plbs) of Vanda coerulea were cultured on an agar medium consisted of halfstrength Vacin and Went (1949) macronutrient, half-strength Murashige and Skoog (1962) micronutrient and 150 ml/l coconut water. After 7 days of culture, plbs were overlaid with 5 cm paraffin-oil or embedded under 1 and 2 cm of water agar medium. The cultures were then incubated either in dark or under 17-25 mol/ m²/s of continuous light from "Day light" fluorescent lamps at 26±2°C. After 168 days of preservation, the plbs embedded under 2 cm agar gave the best result both in term of their appearance and their ability to regrow. This is the first report on agar-embedding conservation in vitro.
