In vitro callus induction from hypocotyl explants of tomato (Lycopersicon esculuentum Mill.) cv. Sida

Abstract

Tomato (Lycopersicon esculentum Mill.) is one of the most important vegetable crops in the world and has been widely grown in almost every country. The effect of growth regulators on callus induction in tomato cv. Sida has been developed. Hypocotyl explants were cultured on Murashige & Skoog (MS) medium supplemented with various concentrations of 6-benzyl aminopurine (BAP) and naphthalene acetic acid (NAA). Effects of plant growth regulators were studied on days for callus induction, percentage of callus induction and callus fresh weight after 4 weeks. The result showed MS medium supplemented with 2.22 µM BAP and 8.06 µM NAA was optimum for callus induction, based on highest percentage of callus induction, highest callus fresh weight and minimum days for callus induction. Hypocotyl-derived callus was successfully maintained and proliferated when subcultured on MS medium supplemented with BAP and NAA. The results provide a basis for efficient micropropagation and genetic improvement of tomato cv. Sida.
