Evaluation of Hom (Strobilanthes spp.) Genotypes Using Molecular Techniques for In Situ Conservation in Phrae Province

Abstract
This study aimed to identify species and assess genetic diversity of indigo plants (Hom) in Phrae province by integrating botanical traits with chloroplast DNA sequence analysis of the psbA–trnH region. Six samples collected from four localities were cultivated under controlled conditions for eight months to enable systematic morphological comparison. The results revealed clear differences in leaf color and size, branching pattern, plant height, and floral traits, allowing classification of SP1 - SP2 as Strobilanthes cusia, SP3 as closely related to Baphicacanthus cusia, and SP4 as Strobilanthes auriculata. Sequence analysis of the psbA–trnH region supported the morphological classification by clustering the samples into three distinct groups and showing 97 - 99% similarity with reference sequences in GenBank. These findings indicate that the psbA–trnH marker is effective for species discrimination and genetic relationship assessment in indigo plants. The generated data provides a useful molecular reference for genetic conservation and for future selection of high-quality indigo resources.
