ผลของสารสกัดน้ำจากใบหูกวางต่อเชื้อ Staphylococcus aureus ภายใต้ความแตกต่างของความเข้มข้นและระยะเวลาการสัมผัส

บทคัดย่อ

เชื้อ Staphylococcus aureus เป็นเชื้อก่อโรคที่ผิวหนังในทั้งสัตว์เลี้ยงและมนุษย์ และมีแนวโน้มการดื้อยาปฏิชีวนะของเชื้อแบคทีเรียสูงขึ้น จึงทำให้สารต้านแบคทีเรียจากธรรมชาติได้รับความสนใจ ดังนั้น งานวิจัยนี้จึงมีวัตถุประสงค์เพื่อศึกษาประสิทธิภาพของสารสกัดน้ำจากใบหูกวาง (Terminalia catappa Linn.) ต่อเชื้อ S. aureus โดยศึกษาผลของความเข้มข้นและระยะเวลาการสัมผัสของสารในการฆ่าเชื้อแบคทีเรีย วิธีการทดลอง ได้แก่ การเตรียมสารสกัดน้ำจากใบหูกวางโดยวิธีต้มและแช่ การหาความเข้มข้นของแทนนินในสารสกัดแช่หมัก และการศึกษาระยะเวลาการออกฤทธิ์ในการฆ่าเชื้อแบคทีเรีย จากการศึกษาพบว่าสารสกัดน้ำจากใบหูกวางที่เตรียมด้วยวิธีต้มและแช่มีความเข้มข้นของแทนนินทั้งหมดเท่ากับ 0.76 มก./มล. และมีค่าความเข้มข้นต่ำสุดที่สามารถฆ่าเชื้อแบคทีเรียด้วยวิธี broth macrodilution assay อยู่ที่ 0.35–0.4 มก./มล. ผลการศึกษาระยะเวลาการออกฤทธิ์ของสารสกัดน้ำจากใบหูกวางต่อการลดจำนวนแบคทีเรีย พบว่าที่ความเข้มข้นของแทนนิน 0.35 มก./มล. ทำให้จำนวนโคโลนีของ S. aureus ลดลงอย่างมีนัยสำคัญทางสถิติ จาก 115 โคโลนีที่ 2 ชั่วโมง เหลือ 0 โคโลนีที่ 24 ชั่วโมง (rs = -1.000, p-value < 0.001) และที่ความเข้มข้น 0.4 มก./มล. ให้ผลในการกำจัดแบคทีเรียอย่างรวดเร็ว โดยจำนวนโคโลนีลดลงจนเหลือ 0 ที่ 8 ชั่วโมง (rs = -0.975, p-value = 0.02) ผลการศึกษานี้ชี้ให้เห็นว่าสารสกัดน้ำจากใบหูกวางอาจเป็นทางเลือกจากธรรมชาติที่ช่วยลดการใช้ยาปฏิชีวนะในการรักษาการติดเชื้อ S. aureus ได้
