การสกัดและการทำให้บริสุทธิ์ของพอลิไฮดรอกซีอัลคาโนเอตชนิดสายโซ่กลางที่ผลิตโดย Streptomyces sp. โดยใช้เอนไซม์และระบบสารละลายน้ำสองวัฏภาค

บทคัดย่อ

พอลิไฮดรอกซีอัลคาโนเอต (PHA) เป็นพอลิเมอร์ย่อยสลายได้ทางชีวภาพ สังเคราะห์โดยแบคทีเรีย มีคุณสมบัติคล้ายพลาสติกจากปิโตรเคมี เป็นมิตรต่อสิ่งแวดล้อม วิธีการสกัดแบบเดิมเป็นอันตรายต่อสิ่งแวดล้อม จึงจำเป็นต้องมีวิธีการสกัดที่คุ้มค่าและเป็นมิตรต่อสิ่งแวดล้อม งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาการสกัดและทำให้บริสุทธิ์ของ PHA จากเซลล์แบคทีเรียโดยใช้เอนไซม์และระบบสารละลายน้ำสองวัฏภาค (ATPS) แบคทีเรียที่ใช้ศึกษาถูกแยกจากดินเพาะปลูก และเพาะเลี้ยงในอาหารเลี้ยงเชื้อสังเคราะห์ที่ประกอบด้วยเกลือแร่ชนิดต่าง ๆ โดยไม่มีแหล่งคาร์บอนที่ซับซ้อน และจำกัดไนโตรเจน เอนไซม์สามชนิด (ไลโซไซม์ ปาเปน และไลเปส) ที่ความเข้มข้น 2–10 กรัม/กรัมเซลล์ ถูกใช้ในการย่อยผนังเซลล์แบคทีเรียที่แยกได้ จากนั้นศึกษาการทำให้บริสุทธิ์ของ PHA ด้วย ATPS ห้าระบบ และตรวจสอบองค์ประกอบ PHA ด้วยวิธีแก๊สโครมาโทกราฟี-แมสสเปกโตรเมทรี ผลการวิจัยพบว่า แบคทีเรียที่แยกได้เป็นชนิดแกรมบวกในสกุล Streptomyces griseoincarnatus เอนไซม์ปาเปน (10 กรัม/กรัมเซลล์) มีประสิทธิภาพสูงสุดในการย่อยผนังเซลล์ รองลงมาคือไลโซไซม์ และไลเปส ที่ความเข้มข้นเดียวกัน โดยให้ค่าการลดลงสัมพัทธ์ของการดูดกลืนแสง 81.9%, 34.2% และ 26.2% ตามลำดับ การสกัดมีประสิทธิภาพสูงสุดเมื่อใช้ไลโซไซม์เข้มข้น 10 กรัม/กรัมเซลล์ ย่อยผนังเซลล์ก่อน 8 ชั่วโมง และตามด้วยปาเปนเข้มข้น 10 กรัม/กรัมเซลล์ อีก 12 ชั่วโมง ให้ผลผลิต PHA สูงสุด 87.2% ระบบ PEG6000/K₂HPO₄ เป็น ATPS ที่มีประสิทธิภาพในการทำให้ PHA ที่สกัดได้มีความบริสุทธิ์เพิ่มขึ้น 58.8% และองค์ประกอบของ PHA ที่สกัดได้เป็นชนิดสายโซ่กลาง ดังนั้นการพัฒนาวิธีการสกัดโดยใช้เอนไซม์และวิธีการทำให้บริสุทธิ์โดย ATPS เป็นอีกแนวทางหนึ่งเพื่อนำไปใช้ในการกู้คืน PHA จากเซลล์แบคทีเรียที่มีประสิทธิภาพและเป็นมิตรกับสิ่งแวดล้อม
