Investigation of toxicity, acetylcholinesterase expression, and digestive tract histopathology in golden apple snails (Pomacea canaliculata) exposed to 2,4-D dimethylammonium herbicide

Abstract

This study aimed to investigate the morphological, biochemical, and histological effects of the herbicide 2,4-D dimethylammonium on the golden apple snail (Pomacea canaliculata). The assessment was based on mortality rates, acetylcholinesterase (AChE) enzyme activity, and histological changes in the digestive tract following exposure to different concentrations of the chemical for 24, 48, 72, and 96 hours. The results revealed that cumulative mortality increased with both concentration and duration of exposure. The LC50 values at 24, 48, 72, and 96 hours were 2,821.95, 2,218.20, 1,651.24, and 1,218.98 µL/L, respectively. A significant reduction in AChE enzyme activity was observed with prolonged exposure, and a decrease in the expression of the 71 kDa AChE protein was detected through SDS-PAGE and Western blot analysis. Furthermore, histological examination revealed various alterations including faded C corpuscles (C cps), darkened K corpuscles (K cps), lumen distention (DL), lamellar collapse (LC), reduction and deformation of corpuscles (RDC), and faded pigmentation (FP). These findings indicate the effects of prolonged and high-concentration exposure to the herbicide at both the molecular and tissue structural levels. From all the data, it is obvious that herbicide 2,4-D dimethylammonium affects golden apple snails. Therefore, application of this herbicide should be carefully monitored to prevent contamination in aquatic environments and bioaccumulation in golden apple snails, which may pose potential health risks to humans through consumption of the snails.
