Effect of 2,4-dichlorophenoxyacetic acid on callus induction of Gymnocalycium mihanovichii for In vitro propagation Effect of 2,4-D and propagation of Gymnocalycium

Abstract
Seeds of Gymnocalycium mihanovichii were surface sterilized by 70% ethanol for 1 minutes followed by 10% Clorox® for 10 minutes and three rinses. The sterilized seeds were culture on Murashige and Skoog (MS) medium for 10 weeks until the seeds germinated into the micro shoots. Then the micro shoots were cultured on MS medium modified by supplementation with 0, 1, 2, 3 and 4 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D). For induction of callus and subsequent embryogenesis, culture had to be incubated at 25±2°C under a 16 hours photoperiod (2,500 Lux) for 3 months. The results showed that, the calli were produced 40.6, 73.6, 75.8, 77.2 and 78.4%, and callus weights of 0.26, 0.49, 0.82, 1.43 and 1.85 g, respectively.
