Effects of Bamboo Mushroom Extract Supplementation with Extenders on Swine Semen Quality in Storage at 17 oC

Abstract

The objective of this study was to determine the effects of bamboo mushroom extract supplementation on semen quality stored at 17 oC. Semen samples were collected from two landrace boars. The ejaculates were diluted in Modena extender with bamboo mushroom extract. The experimental group was divided into a control group without bamboo mushroom extract supplementation (TR1) and a group with bamboo mushroom extract supplementation at concentrations of 10 (TR2), 100 (TR3), and 1,000 (TR4) µg/ml. The quality of sperm was evaluated before the storage on 0, 1, 3 and 5 days by a computer-assisted semen analyzer (CASA). The samples were stored in a cold storage cabinet at 17 °C. The semen quality was assessed by a CASA including total motility, progressive motility, and sperm kinematics.
The acrosome integrity was also assessed using imaging flow cytometry. The results showed that in cold storage at 17 oC, when the sperm was stored on day 5 with a group bamboo mushroom extract at 10 µg/ml had an increase in total motility (88.11±0.38 %) than all experimental groups (P<.05). The bamboo mushroom extract at 10 µg/ml had an increase in curvilinear velocity (85.99±4.84 µm/s), straight line velocity (29.83±4.96 µm/s) and average path velocity (37.43±5.26 µm/s) than all experimental groups (P<.05). Also, the bamboo mushroom extracts 10 µg/ml on the fifth day of semen storage. The percentage of live sperm (75.10±1.24 %) and dead sperm (24.70±1.24 %) was significantly compared to the experiment group (P<.05). Therefore, bamboo mushroom extract supplementation at 10 µg/ml
had a positive effect on swine semen quality in storage at 17 oC.
