Effect of different bicarbonate concentrations on growth performance, carotenoid accumulation and ß–carotene isomer content in Thai Dunaliella salina NUAC09

Abstract

Dunaliella is an important green microalga for the production of natural ß–carotene pigments, which are the economically important substance and high effective radical scavenging capacity. Hence, there are several applications of these pigments in various industries such as nutritional supplements, pharmaceutics and cosmetics. This study was focused on investigating growth performance, accumulations of pigments and cis/trans–ß–carotene isomers ratio of Thai D. salina NUAC09 under different bicarbonate (HCO3) concentrations (0.000, 0.022, 0.043, 0.065 and 0.086 g NaHCO3/L) in Johnson medium. Results showed that the optimum cell density (1.42×106 cell/mL), chlorophyll a content (1.75 µg/mL) and carotenoid content (5.36 µg/mL) occurred in the tested strain cultured at 0.043 g NaHCO3/L. When determined crude extract, the highest total ß–carotene (all–trans and 9–cis) and DPPH radical inhibiting activity found in a strain cultured at 0.086 g NaHCO3/L. Overall, our results indicated that bicarbonate concentration affected growth, pigment content and ß–carotene isomer content of Thai Dunaliella salina NUAC09.
