Effects of different semen extender and storage duration on boar semen quality at 17°C

Abstract

The objective of this study was to investigate the effects of different semen extender formulations in combination with storage duration on boar semen quality. Semen was collected from two boars, five ejaculates per boar, and diluted with extenders. The samples were stored at 17°C for 7 days. The experimental design was a factorial arrangement in a randomized complete block design (RCBD), with two main factors: extender type (BTS, A, and B) and storage duration (1, 3, 5, and 7 days at 17°C). Semen quality was evaluated in terms of motility, abnormalities, and sperm kinematic parameters using a computer-assisted semen analysis (CASA) system. The results showed a statistically significant interaction between extender type and storage duration on progressive motility and several kinematic parameters, including DAP, DSL, DCL, VAL, VCL, STR, and LIN (P<0.05). Semen samples diluted with BTS exhibited significantly higher total motility, progressive motility, VAP, VSL, and VCL across all storage days (1, 3, 5, and 7) compared to the other groups (P<0.05). Samples in Extender A showed higher DSL, DCL, and ALH, while Extender B resulted in higher values of Distal Droplet, STR, LIN, and WOB (P<0.05). In addition, storage duration alone significantly affected total motility and progressive motility, with day 1 showing the highest values (P<0.05). Day 5 was associated with higher values of DMR, DAP, DSL, STR, and LIN compared to other storage times (P<0.05). On day 7, the percentage of sperm abnormalities, particularly distal droplets, was significantly higher than on other days (P<0.05).
