Selection of purple rice lines for fragrance and high yield in F5 to F7 generations between advanced purple rice line and Pathum Thani 1 rice varieties

Abstract

Purple rice is a local rice variety of Thailand that contains several important bioactive compounds which help reduce the risk of various diseases. However, it lacks fragrance, a characteristic highly valued by consumers. Therefore, the improvement and development of purple rice with fragrance is economically and commercially important. In addition, purple rice is photoperiod-sensitive, can be cultivated only once a year, has tall plant stature, and produces relatively low yields compared with modern improved varieties that are photoperiod-insensitive, short-statured, and high-yielding. This research aimed to develop purple rice breeding lines with fragrance and high yield. An advanced purple rice line (KDK10) which is photoperiod-insensitive, short-statured but no fragrance was


crossed with the modern variety Pathum Thani 1 (PTT1), which is fragrant, photoperiod-insensitive, short-statured, and high yielding. Selection for fragrance and yield was conducted from the F5 generation during the wet season of 2021, resulting in nine advanced breeding lines. Further evaluation and selection in the F6 generation during the dry season of 2022 resulted in seven selected lines. In the F7 generation during the wet season of 2022, three promising advanced lines of purple rice were successfully selected. The selected lines exhibited aroma scores of 1.4–1.8, 2-acetyl-1-pyrroline (2AP) contents of 1.0–1.9 mg/kg, and grain yields of 30.7–41.5 g/plant. These advanced purple rice lines show strong potential and will be further evaluated in multi-location trials to confirm their suitability for future varietal development.
