Identification of the fungal fruit rot causal agents of Khiew Sawoey mango of production area in Loei province for export

Abstract

Identification of fungal pathogen causing fruit rot of Khiew Sawoey mango for export in production area of Loei province was conducted, the fruit rot pathogens were isolated, which derived seven isolates of fungi. Pathogenicity tests indicated that all of these fungal isolates were pathogenic on mango fruits. The 7 isolates of fungal pathogens causal agents were identified to general based on morphological characteristics. The result revealed that the isolate KWS1 related to Colletotrichum sp., and isolates KWS4, KWS5, KWS6, KWS12 similar to Lasiodiplodia sp., and KWS7, KWS9 similar to Fusicoccum sp. Molecular identification of those pathogens was performed. The DNA fragment of internal transcribe spacer of rDNA (ITS1-5.8S-ITS2) and translation elongation factor 1-alpha (EF1-α) gene were amplified by PCR technique. The PCR products of 500 and 550 bp were obtained, respectively. Results of nucleotide sequences comparison with GenBank database indicated that the KWS1 was identical to Colletotrichum gloeosporioides, KWS4 and KWS6 to Lasiodiplodia pseudotheobromae; KWS5 and KWS12 to Lasiodiplodia theobromae; KWS7 and KWS9 to Fusicoccum fabicercianum and Neofusicoccum parvum, respectively.
